  重组人粒细胞集落刺激因子在C57BL/6小鼠肠型急性放射病中的作用研究

  摘要:通过建立急性辐射损伤动物模型,研究重组人粒细胞集落刺激因子(rhG-CSF)在C57BL/6小鼠在肠型急性放射病中的作用。将健康C57BL/6小鼠随机分成5组,分别是正常组、10Gy单纯照射组及10Gy 照后rhG-CSF高、中、低浓度给药组。照后观察记录小鼠的生存情况,绘制生存曲线;照后3.5d取骨组织、肠组织观察形态学改变,并对肠组织进行Cleaved Caspase-3阳性细胞计数。实验结果表明:10Gy全身照射后12h给予高浓度rhG-CSF,小鼠生存率提高显著。照后3.5d,rhG-CSF高浓度给药组与10Gy单纯照射组比较,骨组织和肠组织绒毛长度变化不显著,但隐窝凋亡细胞明显减少。提示rhG-CSF可以减少小肠隐窝部位凋亡细胞,在C57BL/6小鼠肠型急性放射病中有一定保护作用。

  Study on effect of rhG-CSF in intestinal form of acute radiation sickness in C57BL/6 mice

  Abstract: To investigate effect of rhG-CSF in intestinal form of acute radiation sickness in C57BL/6 mice by the animal model of acute radiation damage, healthy C57BL/6 mice were randomly divided into 5 gropes, namely the normal grope, 10Gy radiation group, rhG-CSF administration group of high, middle, low concentration after 10Gy radiation . The kaplan-meier survival curve of mice was drew after irradiation; 3.5days after radiation, bone and intestine were taken to observe the pathological change and the number of Cleaved Caspase-3 positive cells were counted. The results showed that 12 hours after 10Gy total body irradiation given the high concentration of rhG-CSF, the survival rate increased significantly. 3.5days after irradiation, comparing high concentrations of rhG-CSF administration group with 10Gy radiation alone group ,bone tissue and the length of intestinal villi did not change significantly, but the number of Cleaved Caspase-3 positive cells in crypts reduced . The results revealed that rhG-CSF reduced the number of Cleaved Caspase-3 positive cells in crypts having a protective role in the intestinal form of acute radiation sickness in C57BL / 6 mice.
