  摘要:组织蛋白酶C(cathepsin C)是免疫细胞中丝氨酸蛋白酶的激活剂,参与人体多种病理生理过程,但是cathepsin C 在中枢神经系统神经炎症中的改变至今未见报道。本研究采用C57BL/6J 小鼠腹腔注射LPS (5 mg/kg),建立神经炎症动物模型。利用免疫组化和原位杂交方法检测注射后脑内小胶质细胞的活化,炎性因子以及cathepsin C 的表达。结果发现,LPS 注射后脑内小胶质细胞活化,炎性因子表达明显,cathepsin C 的表达显著增强,见于多个脑区的小胶质细胞中。上述结果提示,在LPS 诱导的神经炎症中cathepsin C 的表达显著上调,cathepsin C 可能介导了神经炎症反应的发生发展过程,为进一步研究其在神经炎症

  中的意义提供了形态学资料。

  The changes of cathepsin C expression in LPS-induced neuroinflammation in mouse brain

  Abstract: Cathepsin C functions as a central coordinator for activation of many serine proteases in inflammatory cells. cathepsin C is responsible for many inflammatory diseases. In this study,C57BL/6J mice were injected with lipopolysaccharide (LPS, 5 mg/kg i.p.) or saline. Immunohistochemistry (IHC) and In situ hybridization(ISH) methods were used to examine the activation of microglia, the expression of inflammatory cytokines as well as the expression pattern

  and cellular localization of cathepsin C in the brain. The results showed that after LPS injection,microglia was activitated, inflammatory cytokines were expressed significantly, and cathepsin Cwas expressed in microglia in a global pattern. The results indicate that cathepsin C could participate in neuroinflammation induced by LPS.
